pT7-21

The four T7 RNA polymerases are a family with different activity, this T7 RNA polymerase have approximate relative activity of 21% compare to the wild type pT7. We design it as a part of our T7 protein expression device, which is high level and need low-activity promoter to exclude bacterial stress responses. Users can replace the T7 promoter with it or other promoters through the restriction site we added.

Experiment

We expressed GFP on this device in BL21(DE3). Three monoclonal colonies were selected and culture until OD600 0.5(37℃, 220rpm).We calculated the Fluorescence(excitation wavelength-584 nm and emission wavelength-607 nm) and OD600.
